Hey fellow bronies, I'm the one that commissioned the artist (Ajin) to draw that comic last weekend. The glue has no sinister meaning at all, I had completely forgotten that glue was once been made from animals long ago. Luna is simply playing a prank on her sister. Celestia is most definitely calling for Pinkie Pie to help her get revenge, but it sounds much more dramatic to call her "the pink one". Ajin came up with this particular prank, I just asked him to draw Luna pranking Celestia.
I'm thinking of asking Ajin to draw more comics of Luna's pranks. My next idea is to have Luna sneak some pink dye into Celestia's shampoo, and have Celestia come out of the shower dyed pink like her toys. Which will piss off Celestia to no end, of course. I'd love to hear your ideas too. Let me know what pranks you thing Luna should pull on some unsuspecting pony!
On glue: I run with the horse-based-instead-of-synthetic products gags just because of the vague tech levels of Equestria (Electricity having been discovered but still not having largely replaced candles, hoof-held cameras, etc). And for the lulz.
On cutie marks: I choose to believe that Cherilee was just BSing for effect during that lecture. Seriously, ponies would need to have interpreted cutiemarks after they are obtained, so the level of detail behind them is really subjective, not pre-planned (like it could be with a tattoo.
tl;dr -Glue is made of PONIES (and lulz) -Cherilee == Champion BSer
I can't be the only one to figure out Twilight's cutie mark -- that is, the 5 stars (her 5 element of harmony friends) around the center symbol (herself as element of magic).
That would make Twilight's cutie mark a bit different from the others in that it doesn't show a special talent -- it showed a 'prophecy'.
I always though that the Apple family were a bit screwed over with their cutie marks. All they have is fruit which, when compared to the likes of Rainbow Dash's or Twilight's, is kind of lame. Though I suppose its better than having a pitchfork or a ho as your cuite mark.
Funny, I came up with a completely different interpretation of the punchline for the first comic. I thought Pinkie Pie had put Luna up to it (teaching her some pranks), Celestia found out, and was summoning "the pink one" to face some divine justice.
Uh.. Glue is made of hoof material.. well Centuries of having Celestia's hooves being manicured and shaped and shod shoud provide enough clippings to make all the glue needed.

I also object to the reference to That Damn Fanfic in panel 2. Unless that's not really what it is, and the rest of her speech continued as follows:
"...but I think he failed me on purpose so one day I invited him to my place for cupcakes and he said they were so delicious he gave me half a point extra so I got a D- and I was so happy I threw a party and invited all my friends! ...Wait, what were we talking about again?"
I actually don't think they make glue out of ponies in Equestria... What if...and bare with me here...what if it's the other way around there? What if glue is made from HUMANS?! D:
